Differential gene expression analysis between vitis vinifera l. cultivar 'yinhong' and its mutants

    • In order to identify functional genes associated to early-ripening, aroma and peel color; the digital gene expression (DGE) profiles of the grape cultivar Vitis vinifera L. ?Yinhong? and its mutants (YH-20-1 and YH-20-2) were studied. The DGE libraries were constructed from the leaves of ?Yinhong? and its mutants, and then sequenced by the Illimina/Hiseq technique. Field observations and time-intensity (Osme) experiments were also carried out to record the main traits and characters as well as to evaluate the aroma intensity of mature fruit. The results showed significant differences in leaf and fruit traits between ?Yinhong? and its two mutants, under field conditions. Compared to the parent ?Yinhong?, the mutant YH-20-1 showed 3-lobed and deeper leaf blade, and purple black fruit skin; while the mutant YH-20-2 had pink fruit skin, denser leaf pubescence, and early-ripening. The time-intensity method identified a strong fragrance found in YH-20-1 as compared to YH-20-2 and ?Yinhong?. The DGE analysis showed the expression of early-ripening (GSVIVT01015220001 and GSVIVT01011638001) and aroma formation (GSVIVT01011944001, GSVIVT01036445001, GSVIVT01009752001, GSVIVT01009754001, GSVIVT01014569001 and GSVIVT01012363001) related genes were significantly up-regulated in YH-20-1; while in YH-20-2, only an early-ripening related gene (GSVIVT01015220001) was significantly up-regulated. There was no significant difference in aroma formation related genes. Moreover, it was found that resistance related genes (ERF, WRKY family and STS) and neutral amino acid related genes were also up-regulated. Our study suggests that both mutants (YH-20-1 and YH-20-2) were promising with stronger resistance than parent ?Yinhong? and early-ripening by showing purple black fruit skin and pink fruit skin, respectively.
