The incidence and prevalence of malaria presents a much worrying trend and seems to negate the global efforts to control malarial infection. This is in part attributed to a smaller number of antimalarials agents as well as emergence of resistance to the most available antimalarial drugs. Hybrid approach encompassing rational antimalarial drug design involving covalent linking of two pharmacophores into a single molecule has come up as an attractive tool, which could offer advantages such as increased (dual) activity, dosage compliance, countering resistance etc. In this chapter, we have presented update on pyrimidine-based hybrid antiplasmodial agents.
