Fibro nec tin (Fn ) ma trix asse mb ly is a dynamic cellular process in whi ch the soluble dimeric Fn molecules are assembled into insoluble, d isulfide bond stabilized fi brill ar polymeric matrix . Fn matrix assembly req uires s pec ific Fn binding integrin s . Seve ra l Fn binding integrins that are capable of mediating Fn matrix assembl y have bee n ident ified . They include a5B I , a ll bB3 and avB3 integrins. Cells regul a te the matri x assembly process no t o nl y by controlling cell surface expression level of the Fn binding integrins but also by mo dul a tin g F n bindin g a nd cy tos ke le to n b indin g ac ti vities of the integrins. A majo r cha lle nge of future studies is to de lineate the signa l transductio n pathway that regulates Fn matrix assembly.
