Resumen de Perfil electroforético bidimensional de proteínas de la membrana espermática de toros Sanmartinero

  • This study was conducted to analyze the bovine sperm membrane proteins composition using two-dimensional polyacrilamide gel electrophoresis (2D-PAGE) with a polyacrilamide linear gradient gel and the correlation between specific protein and semen characteristics. Semen samples from 10 Sanmartinero bulls, 3 to 4 yrs of age, were assessed for semen quality in terms of spermatozoa motility viability and sperm concentration. Spermatozoa extracts were separated using 2D-PAGE followed by staining with sypro ruby. Three ejaculates per bull were analyzed. Semen was analyzed for percentages of motility and viability and for sperm concentration. In the present work 2DPAGE of proteins extracted from Sanmartinero bull spermatozoa led to the resolution of 485 protein spots, with pIs ranging from 3.3 to 9.5, and molecular weight from 10.5 to 163.6 kDa. Ten protein spots were negatively or positively correlated with some semen quality parameters (motility, viability, concentration). Thus, this study indicates that bovine sperm membrane contains specific proteins that are associated with semen characteristics. It may be that these protein patterns can be used as markers for improving bovine reproduction. The present data provide new and important information on reproduction Colombian creole cattle bull and opens up the possibility to perform additional experiments.
