The oxidative stress induced by reactive oxygen species (ROS) is one of the causes of sperm cryopreservation injury. It has been shown that in vitro capacitation results in increased ROS levels, which are dependent of the antioxidant defense system that can use the NADPH generated by the pentose phosphate pathway (PPP). However, the functioning of the PPP has not been demonstrated in ram spermatozoa, although its involvement in sperm capacitation has already been shown in mouse spermatozoa. The aim of this study was to investigate whether the PPP is operative in ram spermatozoa. We have evidenced the presence of G6PDH and 6PGDH in ram spermatozoa by western blotting, indirect immunofluorescence and RT-PCR. Additional assays to evaluate possible changes in the activity of these enzymes during in vitro capacitation are currently in progress.
