Concentração espermática e fertilização artificial de ovócitos do híbrido tambacu (Colossoma macropomum x Piaractus mesopotamicus)

    Our objective was to determine the best concentration of sperm fertilizing hybrid tambacu (C. macropomum x P. mesopotamicus), a female with advanced maturation, bulging abdomen and reddish genital papilla and a male pacu with the release of semen through light massage being selected abdominal in cranial-caudal direction. The female and the male were induced to play with pituitary extract: Female (1st dose of 0.5 mg.kg-1 and 2nd dose 5 mg.kg-1) and male (single dose of 3 mg.kg-1). A female released in 1560 containing 654 g of oocytes oocytes / g with a rate of 170 040 oocytes / kg, the oocytes were divided into five treatments and three replications: C1 = 1 g, C2 = 1.5 g, C3 = 2 g, C4 = 2.5 g and C5 = 3 g. The male semen produced with 2 mL 10x109 sperm.mL-1, more than 90% motility. Fertilizer was used 20 μL of fresh semen on each repetition. The best ratio sperm / oocyte was found in C4 To better fertilization rate hybrid tambacu is recommended 1 mL of semen pacu for every 125 g of oocyte tambaqui.     Objetivou-se com este trabalho determinar a melhor concentração espermática para fertilização dos ovócitos do híbrido tambacu (C. macropomum x P. mesopotamicus). Para isso, foram selecionadas duas fêmeas de tambaqui com maturação avançada, abdômen abaulado e papila genital avermelhada e quatro machos de pacu com liberação de sêmen mediante leve massagem abdominal no sentido crânio-caudal. As fêmeas e os machos foram induzidos à reprodução com extrato bruto de hipófise. As fêmeas liberaram em média 654 ± 182 g de ovócitos, sendo que cem gramas continham 1560 ovócitos, com uma razão de 170.040 ovócitos.kg-1. Os ovócitos foram distribuídos em cinco tratamentos e três repetições: C1 = 1 g, C2 = 1,5 g, C3 = 2 g, C4 = 2,5 g e C5 = 3 g. Os machos produziram em média 2 ± 0,3 mL de sêmen com 10x109 espermatozóides.mL-1 e média de 90% de motilidade. Para fertilização, utilizou-se 20 μL de sêmen fresco em cada repetição. A melhor relação espermatozóide/ovócitos foi encontrada no tratamento C4. Para melhorar a taxa de fertilização do híbrido tambacu recomenda-se 1 mL de sêmen de pacu para cada 125 g de ovócito do tambaqui.
